Cytokine Receptor IL-12RB2 Gene Polymorphisms Associated with Immune Response to Mumps Viral Vaccine

Abstract

Background: Mumps is described as a highly infectious disease of childhood caused by Mumps virus, the most common symptomslike fever, headache and myalgia are present, also characterized mostly by parotitis and orchitisin some cases. Mumps virus is transmitted by droplets, personal contact and fomites.


Objective: To assess the immune response of school aged children to Measles, Mumps and rubella vaccine(MMR) and study the association of SNP in Interleukin 12 receptor Beta 2 subunit (IL-12RB2) gene with the immune response to MMR.


Methods: A cross-sectional study was conducted on 180 school aged (6-8 years) hospitalized children, previously vaccinated with one or two doses of Measles, Mumps and Rubella (MMR) vaccine. Venous blood samples were collected for detection of immune response level by using enzyme linked immunosorbent assay, followed by conventional polymerase chain reaction for a segment of IL-12RB2 gene and sequencing for detection of SNP.


Results: From 180 Participant (56.2%) males and (43.8%) were females included in the study. Heterogeneous genotype of the rs2201584SNPproduced a significant low immune response in (66.7%) of patients carrying this specific genotype(GA). A significant difference was observed in patients carrying the homogenous mutant genotype (GG) of rs775145846 SNP as they had low immune response.


Conclusion: SNP in IL-12RB2 gene (rs2201584) may play a role in decreasing immune response to Mumps vaccine among school and pre-school aged children. SNP in IL-12RB2 gene (rs775145846) may play a role in decreasing immune response to Mumps vaccine (MuV) among school and pre-school aged children.
